ATCC Medium: 2511 M9 Minimal Agar/Broth
For Solid Media M9 salts
Na2HPO4……………………………..12.8 g
KH2PO4……………………………….3.0 g
NaCl…………………………………..0.5 g
NH4Cl…………………………………1.0 g
DI Water………………………………478 ml
Dissolve M9 salts and autoclave at 121?C.
Agar Solution
Agar…………………………………..15 g
DI Water………………………………500 ml
Heat to dissolve agar and autoclave at 121?C. Once both M9 Salts and Agar Solution have cooled to 50?C, aseptically combine both.
Add the following filter sterilized solutions:
Glucose (20% Solution)……………………..20 ml
1 M MgSO4 Solution…………………………2 ml
1 M CaCl2 Solution…………………………..0.1 ml
Thiamine (0.5% w/v Solution)………………0.1 ml
Dispense complete medium into appropriate vessel.
For Broth M9 salt
s Na2HPO4……………………………..12.8 g
KH2PO4……………………………….3.0 g
NaCl…………………………………..0.5 g
NH4Cl…………………………………1.0 g
DI Water………………………………478 ml
Dissolve M9 salts and autoclave at 121?C. Once both M9 Salts has cooled to 50?C, aseptically combine both.

1 M CaCl2 Solution…………………………..0.1 ml
