FeSO4 . 7H2O .............................5.0 mg
0.04% Phenol red aqueous solution........1.0 ml
Sodium thioglycollate....................0.5 g
Distilled water..........................1.0 L
Adjust pH to 7.6-8.0. For semi-solid and solid medium add 2.0 g or 20.0 g agar respectively to the medium.
In preparing the medium, suspend the uric acid in almost the full volume of liquid, heat to boiling and adjust to a permanent orange color of phenol red (pH 7.6) with NaOH.
Uric acid is relatively insoluble and reacts slowly with alkali in the cold. Autoclave at 121C for 15 minutes.
