Comments注释  This isolate was deposited to ATCC after five passages in weanling mice and two passages in suckling mice. It was adapted to growth in cell culture by three passages in Vero cells (ATCC® CCL-81™).

Growth Conditions生长条件
Temperature培养温度: 37℃
Recommendations for Infection: Plant cells 24 hours in advance and infect when cultures are 80-90% confluent. Remove medium and rinse cell sheet with 1X DPBS.
Remove 1X DPBS and then inoculate with a small volume (e.g. 1 mL per 25 cm2) of virus diluted to provide a MOI of about 0.01. Adsorb for 1 hour at 37℃ in a humidified 5% CO2 atmosphere.
End adsorption by adding virus growth medium. Incubation: Incubate infected culture for 1 to 4 days at 37℃ in a humidified 5% CO2 atmosphere, until CPE are well advance through 90% of the culture. Effect on Host CPE in the form or refractile rounding and sloughing.

EMCV可感染多种哺乳动物、鸟类、昆虫和人类,对多种实验动物宿主有高度传染性。啮齿动物感染EMCV,没有明显的病理变化。自然条件下,EMCV感染最广泛和最严重的动物是猪,感染后可造成突然死亡和实质器官的广泛病理损伤,母猪感染还可造成繁殖障碍。

EMCV在分类上属小RNA病毒科,心肌病毒属,是一种无囊膜的单股正链RNA病毒。病毒粒子直径27nm,20面体对称,外表光滑呈球形。蛋白衣壳由60个衣壳粒子组成,每个衣壳粒子含有4种结构蛋白,即VP1、VP2、VP3和VP4。其中VPl的抗原性最强,且与病毒粒子表面的拓扑结构、抗原性、受体吸附以及脱壳有关。
