空肠弯曲杆菌Campylobacter jejuni subsp. 的保藏ATCC菌种

空肠弯曲菌(ATCC菌种）菌体轻度弯曲似逗点状，长1.5～5 μm,宽0.2～0.8 μm。菌体一端或两端有单鞭毛，运动活泼，在暗视野镜下观察似飞蝇。无荚膜，不形成芽胞。微需氧菌，在含5% O2或10% CO2和85% N2的环境中生长最好。最适生长温度42℃。在正常大气或无氧环境中均不能生长。对于空肠弯曲菌的保藏可大致分为两个第一个是长期，第二个就是短期：
一、长期保藏
1、接种于装有10ml酸水解酪素-酵母浸膏-氯化钠肉汤的试管中，于41℃?44℃培养48±2h。
2、在培养物内加入甘油，使最终浓度达到10%~20%。
3、分装2ml?3ml，于-70℃或更低温度进行冷冻。
4、每6个月传代一次。
二、短期保藏
1、接种于装有10ml酸水解酪素-酵母浸膏-氯化钠肉汤的试管中。
配制方法：20g酸水解酪素，6g酵母浸膏，2.5g氯化钠和8.17g无水酸氢二钾，补足蒸馏水至1L。121℃，15min灭菌。调整pH，使其在灭菌后为8.5±0.2。
2、将接种过的培养基在41℃?44℃培养48h±2h。
3、在巧克力琼脂或血琼脂斜面上进行划线，41℃?44℃培养48±2h。
4、每周进行传代。
注：所有的培养过程必须使用微需氧的空气环境：氮气占85%，二氧化碳占10%，氧气占6%。
