Autoclave at 121? C.
Cool base medium to 45-50?C and add:
Sterile Defibrinated Rabbit Blood…………….……………….150 ml
Broth Medium:
Glycerol………………………………………………………….10 ml
Proteose Peptone………………………………………………10.0 g
Bordet-Gengou Broth Base (see below)……………………..840 ml
Adjust for pH 6.7 +/- 0.2.
Autoclave at 121? C.
Cool base medium to 45-50?C and add:
Sterile Defibrinated Rabbit Blood…………….……………….150 ml
Bordet-Gengou Broth Base Potato…………………………..125 g
NaCl……………………………5.5 g
DI Water………………………..1000 ml
Cut and boil 125 g of potato.
Filter potato broth through cheese cloth and disgard leftover potato.
Clarify broth further by running it through a filter again.
Bring volume back to 1000 ml.
