KH2PO4 ................................................1.0 g
MgSO4 . 7H2O ..........................................0.2 g
Hemin (Sigma H2250) (0.1% in 0.05 N NaOH)............15.0 ml
Distilled deionized water.............................1.0 L
Adjust pH to 6.6 with NaOH. Autoclave 15 minutes at 121C.
Cool to 50C and aseptically add the following filter-sterilized components:
20% Bovine Serum Albumin Fraction V (Sigma A-9647)...10.0 ml
50% Glucose...........................................1.0 ml
10% Cysteine (free base).............................10.0 ml
For liquid medium, omit the corn meal agar and change the phosphate concentration to KH2PO4, 1.5 g and K2HPO4, 0.5 g.
