Yeast extract............................1.0 g

Mineral Solution (see below)............20.0 ml

Trace Metal Solution (see below)........10.0 ml

Distilled water........................970.0 ml

Adjust to pH 7.3 with HCl.

Add:

NaHCO3 ...................................3.0 g

0.1% Resazurin...........................1.0 ml

Boil under 80% N2, 20% CO2; dispense and seal under same gas phase. Replace headspace with same gas phase at 1 atm over pressure. Autoclave at 121C and cool.

Add aseptically:

Vitamin Solution (see below)............10.0 ml

Sodium pyruvate.........................40.0 mM  

50 mM Na2S2O4 (freshly prepared).........10.0 ml

Mineral Solution:

NaCl....................................40.0 g

NH4Cl ...................................50.0 g

KCl......................................5.0 g

KH2PO4 ...................................5.0 g

MgCl2 . 6H2O .............................8.3 g

CaCl2 . 2H2O .............................1.0 g

Distilled water..........................1.0 L

Trace Metal Solution:

Nitrilotriacetic acid....................2.0 g

MnSO4 . H2O ..............................1.0 g

Fe(NH4)2(SO4)2 . 6H2O .....................0.8 g

CoCl2 . 6H2O .............................0.2 g

ZnSO4 . 7H2O .............................0.2 g

CuCl2 . 2H2O .............................0.02 g

NiCl2 . 6H2O .............................0.02 g

Na2MoO4 . 2H2O............................0.02 g

Na2SeO4 ..................................0.02 g

Na2WO4 ...................................0.02 g

Distilled water..........................1.0 L

Add nitrilotriacetic acid to water and adjust to pH 6.0 to solubilize prior to addition of remaining components.

2 Vitamin Solution:

Pyridoxine hydrochloride.................5.0 mg

Riboflavin...............................5.0 mg

Calcium D-(+)-pantothenate...............5.0 mg

p-Aminobenzoate..........................5.0 mg

Thioctic acid (in 0.1 N NaOH)............5.0 mg

Nicotinamide............................50.0 mg

Biotin...................................5.0 mg

Cyanocobalamin...........................5.0 mg

Hemin (in 0.1 N NaOH)....................5.0 mg

1,4-Naphthoquinone (in 0.1 N NaOH)......20.0 mg
