Basal Medium:
NaCl.......................250.0 g
MgSO4 . 7H2O ................20.0 g
Trisodium citrate............3.0 g
KCl..........................2.0 g
Tryptone.....................5.0 g
Yeast extract................3.0 g
Distilled water..............1.0 L
Autoclave at 121C for 15 minutes.
Add 0.1 ml of Trace Metals (See below) to sterile basal medium. Dispense aseptically.
Trace Metals:
ZnSO4 . 7H2O .................1.32 g
MnSO4 . H2O ..................0.34 g
Fe(NH4)2SO4 . 6H2O ............0.82 g
CuSO4 . 5H2O .................0.14 g
Distilled water............200.0 ml
Filter-sterilize.
Agar Medium:
For agar medium, dissolve the ingredients for the basal medium in 600 ml of water.Autoclave the basal medium at 121C for 15 minutes.
In a 2.0 liter flask, dissolve 20 g of agar in 400 ml of distilled water.Autoclave at 121C for 15 minutes.
Combine the sterile basal medium and this agar solution; add 0.1 ml of the Trace Metals solution. Dispense as required.
