DI Water……………………………………….900.0 ml
Prepare separately each of the following stock solutions and add to the basal medium as indicated below to avoid precipitation. (Make final volume to 1.0 L)
Stock Solutions: 0.05M CaCl2…………………………………..8.0ml
0.4 M MgSO4 x 7H2O………………………..10.0 ml
0.25 M Na2HPO4 x 7H2O………………...….10.0 ml
0.25 M KH2PO4……………………………….10.0 ml
Na Citrate x 2H2O…………………………….1.0 g
0.005 M Fe(NH4)2(SO4)2 x 6H2O………….10.0 ml
The pH should be 6.5. Autoclave at 121 C. Filter sterilize a 2 M glucose solution and add aseptically 50 ml to the above medium. 18g/50ml glucose = 2M glucose.
