中国微生物菌种查询网白喉棒状杆菌Corynebacterium diphtheriae，菌体大小为（0.3~0.8μm）×（1~5μm），细长稍弯，粗细不一，菌体一端或两端排列呈棒状，排列不规则，常呈L、V、X、T等字形或排成栅栏状。革兰氏染色阳性，无荚膜、鞭毛，不产生芽胞；用美兰液染色菌体着色不均匀，常呈着色深的颗粒；用奈瑟氏染色菌体染成黄褐色，一端或二端染成蓝色或深蓝色颗粒，称为异染颗粒（Metachromatic granules），其主要成分是磷酸盐和核糖核酸，是本菌形态特征之一 。

本菌的致病物质主要是白喉毒素。白喉毒素是含有两个二硫键的多肽链，分子量为62，000。经蛋白酶水解后，可分为A和B两个片段，中间仍由二硫键联接。B片段，无酶活性，但能与宿主易感细胞表面特异性受体结合，并通过易位作用使A片段进入细胞。A片段具有酶活性，能将氧化型烟酰胺腺嘌呤二核苷（NAD+）水解为烟酰胺及腺嘌呤二磷酸核糖（ADPR）两部分，并催化延伸因子-2（Elongation factor-2,EF-2）与ADPR共价结合，使EF-2失去转位活性，从而中止肽-tRNA及mRNA在核糖体上由受位转移至供位，肽链不能延长，细胞蛋白质合成受阻，细胞死亡，病变产生。
