常用细胞系配置培养基

 我们单位细胞库建议使用背景资料上指定的培养基培养细胞系，部分常用的培养基配制方法罗列如下，培养基过滤除菌后应检测无菌状况，然后补加血清和双抗，双抗的使用浓度一般是终浓度100单位/mL培养基。当然，部分细胞可用其它培养基替代，每个实验室都有自己传统的培养基配制方法，我们提供的配制方法仅供参考。

 
1.消化液的配制：
①胰酶（1：250）  2.5 克
②EDTA-Na         0.3克
③NaCl            8.0克
④KCl             0.4克
⑤Glucose         1.0克
⑥Phenol Red      0.005克
⑦Water           1000 mL
磁力搅拌2-3小时，调pH 7.8-8.0，过滤除菌。
2．MEM  medium  (1L)
   ① MEM                     1瓶（代）
      Non-essential amino acids     10  mL
      丙酮酸钠                  0.11克
water                      800 mL
磁力搅拌2-3小时
   ②  NaHCO3                            2.0克
water                    200 mL
磁力搅拌1小时
①+②, 调pH 7.0-7.2，过滤除菌。
3. RPMI 1640  medium  (1L)
① RPMI 1640                     1瓶（代）
      L-Glutamine                     0.11克
丙酮酸钠                       0.11克
water                          800 mL
磁力搅拌2-3小时
    ②  NaHCO3                            2.0克
water                    200 mL
磁力搅拌1小时
①+②, 调pH 7.0-7.2，过滤除菌。
 
4. DMEM  medium  (1L)
①  DMEM                       1瓶（代）
 Glucose                       3.5克
water                          800 mL
磁力搅拌2-3小时
    ②  NaHCO3                            2.0克
water                    200 mL
磁力搅拌1小时
①+②, 调pH 7.0-7.2，过滤除菌。
 
5. Grace`s  Medium
    Grace`s Insect Cell Culture Medium              1瓶（代）
        Yeast extract                                 3.3克
         Lactal albumen hydrolysate                    3.3克
         NaHCO3                                            0.35克
water                                     1000 mL
在15-30℃磁力搅拌4-5小时
用2N  KOH 调pH至6.5-6.6，过滤除菌。
 
 6．DMEM/F12 Medium
    ①  DMEM/F12                                  1瓶（代）
L-Glutamine                                  0.2克
丙酮酸钠                                     0.11克
water                                      800 mL
磁力搅拌2-3小时
②  NaHCO3                                             2.0克
water                    200 mL
磁力搅拌1小时
①+②, 调pH 7.0-7.2，过滤除菌。
 
7. 部分常用的微生物检测培养基
7.1 Sabouraud dexrosebroth  (Difco 0382-01)  SDB
   Sabouraud powder    3.00g
   Distilled water       100ml    pH 5.6
   Dispense 3ml aliquots of broth into each of 33 test tubes (12×100mm), and cap each tube loosely.
 
7.2 Brain heart infusion broth BHI (Difco 0037-01-6)
   Broain heart infusion powder   3.70g
   Distilled water               100ml     pH 7.4
   Dispense 33 test tubes (12×100mm), and cap each tube loosely.
 
7.3 Thioglycollate medium THIO (Difco 0256-01)
   Thioglycollate powder     2.98g
   Distilled water            100ml        pH 7.1
   Dispense 33 test tubes (12×100mm), and cap each tube loosely.
 
7.4  Trypticase soy broth TSB (BBL 01-162)
    Trypticase soy broth powder    3.00 g
    Distilled water               100ml    pH 7.3
    Dispense 33 test tubes (12×100mm), and cap each tube loosely.
All medium are sterilized in an autoclave for 20 min at 15 lbc pressure (121℃) on slow exhaust. Store at room temperature or 4℃ 
 
8,包被用多聚赖氨酸配制
取25mg溶于50ml  H2O中，过滤除菌，再取4ml 稀释至50ml H2O中 成工作液，4度保存。
