木醋杆菌的培养条件
材料
木醋杆菌种子
培养基
葡萄糖20g/L，蛋白胨5g/L，酵母粉5g/L，柠檬酸1g/L，PH6.0
仪器
恒温培养摇床,智能生化培养箱,红外光谱仪,扫描电镜,热重分析仪
培养方法
挑取活化好的菌种接入种子培养基，30摄氏度，160r/min,振荡培养24h后以6%的接种量接入液体培养基，30摄氏度下静置培养7天。
样品纯化处理方法
将培养木醋杆菌所得凝胶膜经蒸馏水漂洗，1 %氢氧化钠溶液煮沸4h，于室温下在0.5%乙酸中浸泡0.5H，蒸馏水冲洗烘干后称重。
样品的性能测试
细菌纤维素样品用变换红外光谱仪分析有机阴离子官能团，测试波数范围为4000-400cm。用电子显微镜观察样品的表面形貌。热重分析采用热重分析仪,称取2.031mg粉末，升温速度10摄氏度/min，检测温度25-45摄氏度。
最佳培养环境
葡萄糖30g/L，蛋白胨7.5g/L，酵母粉10g/L，磷酸氢二钠7.5g/L，柠檬酸0.5g/L，PH0.5。 五个因素对细菌纤维素产量的影响次序为：酵母粉>PH值>葡萄糖>柠檬酸>蛋白胨。
