This material is cited in a U.S. and/or other patent and may not be used to infringe the patent claims. Recombinant virus produces focal CPE, which appears as blue plaques when overlayed with X-Gal (150 µg/mL in serum supplemented D-MEM with 0.5% agarose).
Effect on Host CPE, cell enlargement, rounding and degeneration
Recommended Host MRC-5 cells (ATCC® CCL-171™)
Growth Conditions Recommendations for Infection: Infect host cells in a T-25 flask or equivalent culture vessel, at 80-90% confluence. Incubate for 2 to 9 days at 37℃ in a humidified 5% CO2 atmosphere. For passage, co-cultivate infected cells with fresh cells.
For preservation, harvest infected cells by trypsinization, resuspend in culture fluids supplemented with DMSO to 8%, and freeze.
